Change Notification - Reporting Post-Approval Changes to Regulators

A change notification determines how a post-approval manufacturing, formulation, or facility change must be communicated to FDA once an internal change control record has approved the change itself. The regulatory reporting category, Annual Report, CBE-30, CBE-0, or Prior Approval Supplement, is driven entirely by the change's potential impact on product identity, strength, quality, purity, or potency. This map details the pathway from change identification through classification, submission, and implementation.

Annual Report

Minimal potential impact change, reported in the annual report rather than submitted individually.

Impact: Minimal

CBE-30

Moderate potential impact change effected 30 days after FDA receipt, absent an FDA objection.

Impact: Moderate

CBE-0

Moderate potential impact change effected immediately upon FDA receipt of the supplement.

Impact: Moderate, urgent

Prior Approval Supplement

Substantial potential impact change requiring FDA approval before the change may be implemented.

Impact: Substantial
Phase 1
Change Identification and Impact Assessment
1Identify a proposed post-approval change
Change identified and approved through the internal change control system before regulatory reporting is considered.
Internal Change Control
↓
2Assess potential impact on product quality
Change assessed for its potential effect on the product's identity, strength, quality, purity, or potency.
21 CFR 314.70
↓
3Determine the regulatory reporting category
Category determined as Annual Report, CBE-30, CBE-0, or Prior Approval Supplement based on the impact assessment.
SUPAC Guidance
↓
4Consult regulatory affairs for classification confirmation
Proposed classification reviewed and confirmed by regulatory affairs before supporting data generation begins.
Internal SOP
Phase 2
Supporting Data Generation
5Generate supporting data
Stability, comparability, or validation data generated to demonstrate the change does not adversely affect the product.
21 CFR 314.70
↓
6Compile the change notification package
Supporting data, updated CMC sections, and justification assembled into the applicable submission package.
Internal SOP
↓
7Conduct internal QA and regulatory review
Package reviewed by quality and regulatory affairs for completeness and consistency with the determined category.
Internal SOP
↓
Does the change require prior FDA approval before implementation?
YES → Submit PAS and await approval NO → Proceed per CBE or annual timeline
Phase 3
Submission and Regulatory Interaction
8Submit the notification to FDA
Submission filed as a CBE-30, CBE-0, Prior Approval Supplement, or included in the next Annual Report as applicable.
21 CFR 314.70
↓
9Respond to FDA information requests
Follow-up questions or requests for additional data from the review division addressed promptly and completely.
21 CFR 314.70
↓
10Track the applicable waiting period
The 30 day waiting period for a CBE-30 submission tracked precisely, confirming no FDA objection is received.
21 CFR 314.70
Phase 4
Implementation and Closure
11Implement the change following the applicable timeline
Change implemented only once the applicable waiting period has elapsed or FDA approval has been received.
21 CFR 314.70
↓
12Update the site master file and batch records
Internal documentation, batch records, and site master file updated to reflect the approved and implemented change.
Internal SOP
↓
13Receive FDA approval or complete the notification period
Prior Approval Supplement approved by FDA, or the CBE waiting period completes without objection, closing the notification.

Change Reporting Categories (Overview)

Annual Report

Minimal impact changes bundled into the product's next scheduled annual report to FDA.

21 CFR 314.70(d)

CBE-30 / CBE-0

Moderate impact changes effected either 30 days after filing or immediately, depending on urgency and risk.

21 CFR 314.70(c)

Prior Approval Supplement

Substantial impact changes that cannot be implemented until FDA reviews and approves the supplement.

21 CFR 314.70(b)

Change Impact Criteria (Overview)

Minimal potential impact Change has little to no potential to affect the identity, strength, quality, purity, or potency of the product.
Moderate potential impact Change has a moderate potential to affect product quality, but risk is adequately controlled by supporting data.
Substantial potential impact Change has substantial potential to affect product quality, warranting FDA review before implementation.
Classification drives the category The impact level assigned determines whether the change can proceed immediately, after 30 days, or only after approval.

Key Change Notification Outputs (Overview)

Change notification package Complete submission including supporting data, updated CMC content, and category justification.
FDA approval letter Formal approval received for a Prior Approval Supplement before the change may be implemented.
Updated regulatory filing Approved application updated to reflect the implemented change across all relevant CMC sections.
Post-implementation verification Confirmation that the change was implemented as approved and that no unintended quality impact occurred.

Change Notification Quality Elements

Accurate Impact Classification Change consistently classified against the actual potential impact on identity, strength, quality, purity, or potency.
Supporting Data Sufficiency Stability, comparability, or validation data sufficient to support the assigned reporting category.
Cross-Functional Regulatory Review Quality and regulatory affairs jointly review the classification and package before submission.
Timely Submission Notification submitted promptly once supporting data and internal approvals are complete.
Waiting Period Discipline Change implementation withheld until the applicable CBE waiting period or FDA approval is confirmed.
Global Filing Alignment Equivalent notifications or variations filed with other regulators where the same change applies globally.
Post-Implementation Verification Change verified as implemented correctly, with no unintended effect on product quality after the fact.
Documentation Traceability Complete traceability from the internal change control record through to the regulatory submission and approval.

Never do this

Implement a change before the applicable waiting period or approval is confirmed. Classify a change based on convenience rather than its actual potential impact. Submit a notification package without sufficient supporting data to justify the category. Leave a global filing unaligned when the same change applies to other markets.

Change notification vs change control

Change control is the internal quality system process that evaluates, approves, and implements a change. Change notification is the separate regulatory reporting step that communicates an approved change to FDA, with its own classification and timeline requirements distinct from internal approval.

CBE-30 vs CBE-0 vs PAS

A CBE-30 change may be implemented 30 days after FDA receipt absent an objection. A CBE-0 change may be implemented immediately upon receipt. A Prior Approval Supplement cannot be implemented until FDA affirmatively approves the change.
