Process Validation - A Three-Stage Lifecycle, Not a Single Event

FDA's 2011 Process Validation Guidance reframed validation as a continuous lifecycle spanning three distinct stages: Process Design, where the commercial process is defined based on development knowledge; Process Qualification, where the design is confirmed capable of reproducible commercial manufacturing; and Continued Process Verification, where ongoing data confirms the process remains in a state of control throughout commercial production. This lifecycle model replaced the older view of validation as three consecutive successful batches completed once before launch and then left largely unexamined.

Traditional (Prospective) Validation

Process qualification completed and evaluated before the product is released for commercial distribution, the most common approach for new products and processes.

Timing: Before commercial release

Concurrent Validation

Product released for distribution based on monitoring and testing of the specific batches used for qualification, applied only under justified, documented circumstances.

Timing: Justified exception basis

Retrospective Validation

Validation based on accumulated historical production and testing data for an already-marketed process, now largely discouraged in favor of prospective or lifecycle approaches.

Status: Legacy, largely discouraged

Continuous Process Verification

An alternative approach, often PAT-enabled, where process performance is continuously monitored and evaluated throughout commercial manufacture in place of discrete PPQ batches.

Basis: ICH Q8, continuous monitoring
Phase 1
Process Design (Stage 1)
1Define the Quality Target Product Profile and CQAs
QTPP established describing the product's intended performance, with Critical Quality Attributes identified as the measurable properties that must fall within an appropriate range to ensure quality.
ICH Q8
↓
2Identify Critical Process Parameters
CPPs identified through risk assessment and development data as the process inputs whose variability has a direct, meaningful impact on a CQA.
ICH Q8 · ICH Q9(R1)
↓
3Establish the design space and control strategy
Multidimensional combination of process parameters demonstrated to assure quality established where applicable, with an overall control strategy defined based on process understanding.
ICH Q8 · ICH Q10
↓
4Finalize process design documentation
Process design and control strategy formally documented, providing the technical basis for the qualification protocols developed in the next stage.
FDA PV Guidance 2011
Phase 2
Process Qualification (Stage 2)
5Qualify facility, utilities, and equipment
Installation Qualification and Operational Qualification completed for all facility, utility, and equipment elements supporting the process before performance qualification begins.
EU GMP Annex 15
↓
6Execute Process Performance Qualification runs
PPQ batches manufactured under actual production conditions, using qualified equipment, trained personnel, and approved procedures exactly as commercial production would occur.
FDA PV Guidance 2011
↓
7Sample and test extensively during PPQ
Enhanced sampling and testing performed during PPQ runs relative to routine production, providing statistically meaningful evidence of reproducibility.
FDA PV Guidance 2011
↓
Do PPQ results meet the pre-defined acceptance criteria?
YES → Proceed to continued verification NO → Investigate and revise
Phase 3
Continued Process Verification (Stage 3)
8Establish the ongoing monitoring plan
Monitoring plan defined specifying which CPPs and CQAs are tracked routinely, at what frequency, and against what statistical criteria during commercial production.
FDA PV Guidance 2011
↓
9Collect and trend routine production data
Data from every commercial batch collected and trended over time, providing the ongoing evidence base that the process remains in its validated state.
ICH Q10
↓
10Detect and respond to process drift
Statistical signals of process drift or increased variation investigated promptly, even where individual batch results remain within specification.
ICH Q10
Phase 4
Documentation and Change Management
11Compile and approve the validation report
Complete validation report compiled covering all three stages and formally approved by the quality unit before commercial release depends on the validated process.
EU GMP Annex 15
↓
12Evaluate changes through change control
Any proposed change to the validated process evaluated through change control for its potential impact on validated state, with revalidation scope determined accordingly.
EU GMP Annex 15
↓
13Conduct periodic review of validated state
Validated state reviewed periodically, incorporating CPV data, deviation history, and any accumulated process knowledge since the last review.
ICH Q10
Three-Stage Requirements Summary

Stage 1 - Process Design

Establishes the scientific and risk-based foundation the commercial process is built on, using development-phase knowledge and data.

FDA PV Guidance 2011

Stage 2 - Process Qualification

Confirms the process design is capable of reproducible commercial manufacturing, through facility/equipment qualification and PPQ batch execution.

FDA PV Guidance 2011

Stage 3 - Continued Process Verification

Provides ongoing assurance during commercial production that the process remains in a state of control, continuing for the entire product lifecycle.

FDA PV Guidance 2011

PPQ Failure Response Steps

Step 1 - Secure all PPQ data and samples Preserve complete batch records, testing data, and retained samples from the failed PPQ run before further action.
Step 2 - Investigate the specific failure mode Determine which CQA or CPP failed to meet acceptance criteria and at what point in the process the deviation occurred.
Step 3 - Assess process design adequacy Evaluate whether the Stage 1 process design and control strategy require revision based on what the PPQ failure revealed.
Step 4 - Revise and re-qualify as needed Implement necessary process, equipment, or procedural changes and repeat PPQ execution with the revised approach.
Step 5 - Document the full investigation and resolution Maintain a complete record connecting the original failure, root cause, corrective action, and successful re-qualification outcome.

Sampling and Statistical Requirements

Justified PPQ batch number Number of PPQ batches justified based on process complexity and variability, rather than defaulting to a fixed number without rationale.
Enhanced sampling during PPQ Sampling plan during PPQ runs more extensive than routine production sampling, providing the statistical confidence needed to demonstrate reproducibility.
Statistical confidence in conclusions Acceptance criteria and conclusions drawn from PPQ data supported by an appropriate statistical basis, not solely a pass/fail check against specification.
CPV data statistically evaluated Stage 3 monitoring data evaluated using statistical methods capable of detecting meaningful drift, not simply reviewed batch by batch in isolation.

Validation Lifecycle Elements

Quality Target Product Profile (QTPP) Prospective summary of the quality characteristics a drug product should possess to ensure the desired quality, safety, and efficacy.
Critical Quality Attributes (CQAs) Physical, chemical, biological, or microbiological properties that must be controlled within an appropriate range to ensure product quality.
Critical Process Parameters (CPPs) Process parameters whose variability has a direct and significant impact on a critical quality attribute.
Design Space Multidimensional combination of input variables and process parameters demonstrated to provide assurance of quality, where established.
Control Strategy Planned set of controls derived from product and process understanding that assures process performance and product quality.
IQ / OQ / PQ Installation, Operational, and Performance Qualification providing the documented evidence that equipment and systems function as intended.
Revalidation Triggers Defined events, such as significant process, equipment, or raw material changes, that trigger a formal revalidation assessment.
CPV Data Sources In-process data, release testing, stability data, complaint trends, and deviation history feeding into the ongoing Stage 3 monitoring picture.

Never do this

Treat validation as complete once PPQ batches pass, without an ongoing Stage 3 monitoring plan. Set the PPQ batch number as a fixed default without complexity-based justification. Implement a process change without evaluating revalidation impact through change control. Rely solely on pass/fail specification checks without statistical evaluation of PPQ data.

Traditional vs continuous verification

Traditional validation relies on a discrete set of PPQ batches completed before commercial release. Continuous process verification, often enabled by process analytical technology, continuously evaluates process performance throughout commercial manufacture, providing an alternative pathway to the same lifecycle assurance.

Qualification vs validation

Qualification, covering IQ, OQ, and PQ, establishes that equipment and facilities function as designed and intended. Validation is the broader concept establishing that the entire manufacturing process, using that qualified equipment, reliably produces a product meeting its predetermined quality attributes.
