Compounding Animal Drugs: GFI #256 and Draft GFI #256B

When FDA does not intend to take enforcement action on animal drug compounding, and what the new draft changes.

What this course covers

Compounding from bulk drug substances fills gaps where no approved animal drug will do, but sits outside the approval system, so FDA relies on guidance to say when it does not intend to take enforcement action. This course explains that policy for compounders, pharmacists, veterinarians, and quality professionals, including the new draft for federally registered facilities.

The course begins with why compounding needs a policy at all: a drug made from a bulk drug substance is not an approved animal drug, so it can violate the new animal drug approval and adequate directions for use requirements. Learners see how enforcement discretion works.

Next, learners study GFI #256, which covers compounding by or under the supervision of veterinarians and by pharmacists in state-licensed pharmacies or federal facilities. They review the covered circumstances: patient-specific prescriptions and office stock for non-food animals, antidotes for food-producing animals, and sedatives or anesthetics for free-ranging wildlife, with the state law and withdrawal conditions.

The course then covers draft GFI #256B, issued August 27, 2026, which extends the policy to outsourcing facilities registered under section 503B(b) and facilities registered under section 510(b), with registration, listing, and direct distribution conditions.

Learners then focus on where the draft is stricter: CGMP compliance is a condition of eligibility, and facilities that are also state-licensed pharmacies should fully segregate CGMP and non-CGMP operations, labeling included.

Finally, the course covers the draft’s status, the November 27, 2026 comment deadline, and records that support reliance on the policy.

Why it matters

A drug made from a bulk drug substance is not an approved animal drug. FDA’s guidance says when it does not intend to take enforcement action, and a compounder has to meet its conditions to rely on it.
